In physical organic chemistry, the cinetic isotope effect (KIE) is a change in the reaction rate of a chemical reaction when one of the atoms in the reagents is replaced by one of its isotopes. Formally, the ratio of the rate constants for reactions involving light (kL) and heavy (kH) isotopically substituted reagents (isotopologists).

This change in response rate is a quantum mechanical effect, primarily due to the lower vibrational frequency of heavier isotopologists compared to their lighter counterparts. In most cases, this implies a higher energetic input needed for heavier isotopologists to reach the transition state (or, in rare cases, the dissociation limit) and, consequently, a slower response rate.
